The effect of the administration of the actin polymerization inhibitor cytochalasin D during the in vitro capacitation of boar spermatozoa was assessed. In particular the acquisition of fertilizing ability and the main intracellular signalling pathways were analyzed. It was found that treated spermatozoa were unable to fulfil the capacitation, even if the biochemical analyses carried out on signalling machinery failed to point out any biochemical alteration. By using a computational model, based on biological networks theory and representing the molecular interactions involved in male gametes post-ejaculatory life, it was possible to explain these data: the actin polymerization resulted to interact with the different subcellular compartments, acting as a coordinator of molecular pathways involved in sperm cells function.
